Abstract
The sparid fish striped sea bream (Lithognathus mormyrus) was chosen by us as an adequate sentinel organism for bio-monitoring coastal Mediterranean habitats. Using the subtractive-suppression- hybridization procedure, a 4704-fragment fully sequenced and annotated hepatic cDNA transcriptome was cloned from striped sea bream exposed to a variety of pollutants. This transcriptome was used for the construction of a cDNA microarray of duplicate clones which contain 1502 unique sequences, used to identify changes in hepatic gene expression profile upon exposure to cadmium applied by feeding or intraperitoneal injections. Each treatment and the control group included four fish in reproductive arrest. Dual-labeled cDNAs, originated from hepatic transcript populations of individual fish were hybridized to the microarray in a loop design. Each loop contained one fish of each group. The hybridization results were analyzed by the LIMMA and EXPANDER software tools. 32 unique sequences altered their expression levels upon exposure to cadmium. They include 12 non-annotated sequences. Annotated genes include proteases, protease inhibitors, proteins belonging to the immune system and proteins involved in transport and metabolism of transition metals. Expression patterns of individual fish were clustered and a model for using the cDNA microarray as a bio-monitoring tool is suggested.
